This can be accomplished by using the Email Delivery service monitor, which tests sending email through one server and receipt of email on another server.

To test flow between two servers, set up a special user account on the delivery server that will automatically forward inbound emails to another account on the second server. Then configure the Email Delivery monitor to send email to the delivery server and retrieve the email from the second server to confirm that the email made the desired round trip.
